Chromatin is a highly dynamic and complex structure that undergoes changes in response to environmental cues. Even small changes at the atomistic level can lead to large perturbations in the overall chromatin structure and function due its hierarchical organization with many levels of regulation. We study the roles of DNA and histone modifications in precise regulation of chromatin compaction, DNA accessibility and binding of chromatin factors. The transient and intricate nature of chromatin dynamics makes it difficult to study it solely through experimental approaches. Semi-quantitative experimental methods may offer context-specific information but often lack single-nucleotide and atomistic precision. To gain insights into chromatin dynamics at high-resolution, we develop integrative approaches by combining experimental data with atomistic molecular dynamics simulations, machine learning and molecular modelling.
